《幕府将军2》的游戏时代设定在十六世纪的日本战国时代，这个时候的日本被众多大名把持着，各大名之间为了争夺土地和日本的统治权，互相讨伐不断，这给游戏提供了充分的素材，游戏中。你的任务就是扮演一名战国时代的大名，培植亲信，建立军队，通过外交、经济、暗杀等手段，逐步达到你的目的——作为胜利条件，你的目的有四个：消灭你的敌对家族；让你的家族存活七十年；占领日本四十个藩国；统一日本。　　游戏提供了九方势力可选：毛利、武田、织田、北条、岛津、今川、上杉、叛军、足利幕府，每一方都有自己的根据地及军/政强项。其中几家孤悬海外偏安一隅，自给自足但难以干预时政；另一些则整天大小阵仗不断，要靠手下的武士拼死维持局面。
　　
这是一个时局变换莫测的时代，由荷兰及葡萄牙输入的火枪此时尚未登岛，能主宰战场的只有武士们的血肉之躯。游戏专注于战场细节，同屏作战单位通常不超过40个（每方20个左右），每一个单位都有自己明确的目的和用途且提供升级选项。角色级别由甲胄颜色及背后所插旗帜区分，较以往更易分辨。　　除亚洲特有的人物崇拜之外，到处筑墙的习惯也对游戏战役产生了实质性影响。每一座城池都有五层防御体系，甚至筑垒地域也因临海或依山从而拥有某些防御加成。
　　
全战系列中经典的海战场面也将出现在《幕府将军2》中，只不过双方的船都由桨驱动，无需担心风向的影响。海战的主要模式就是双方成堆的弓箭手互射，然后互抛抓勾使双方靠帮从而爆发决定性的接舷战。

以往全战作品所不同的是，首次实现了多人模式下的组队对抗；最多可支持八人游戏，每一方都可使用特定的人物形象，且获得的经验值可为自己所扮演的角色升级。
